Well.... you could start by reading the excellent manual... or even read the lovely digital docs in PDF format.
Everything, down to the last little submenu is described in detail.

....or if you're looking for creative advice on how to achieve realistic paint results, don't think twice about buying one of the Painter WOW! books.
These are a really excellent source, as how to get the most out of painter.
